Progress toward a biosynthetic rationale of the aflatoxin pathway

چکیده
Specifical ly—labeled samples of averufin have been synthesized and incorporated intact into aflatoxin B1, versicolorin A and versiconal acetate as determined by NMR spectroscopy. [1—'3C]--Hexanoic acid has been shown to be uti I ized as the primer of the pathway whi le a group of alternative starters was found to be degraded to acetate prior to incorporation. The correct structure of nidurufin has been established and it and its 2'—epimer, pseudonidurufin, in labeled form have failed to be utilized in aflatoxi n biosynthesis under two experimental protocols. [1 '..l8, 5' —'3C]—Averufi n, however, was observed to label versiconal acetate in such a way that the acetyl carbonyl bore 80% of the original 180—label. A biogenetic scheme is proposed to account for these findings that is supported in the cat ionic regime by preliminary model studies.

Expression of aflR, veA and laeA as regulators of aflatoxins and cyclopiazonic acid biosynthesis pathway in Aspergillus flavus
In this study, the production of aflatoxin B1 (AFB1) and cyclopiazonic acid (CPA) was investigated in toxigenic and non-toxigenic Aspergillus flavus with respect to expression of aflR, veA and laeA genes that are involved to toxins production. A. flavus strains were cultured in YES broth at 28 °C for 4 days and the presence of (AFB1) and (CPA) was confirmed and measured by TLC and HPLC. The exp...
متن کاملCurrent Understanding on Aflatoxin Biosynthesis and Future Perspective in Reducing Aflatoxin Contamination
Traditional molecular techniques have been used in research in discovering the genes and enzymes that are involved in aflatoxin formation and genetic regulation. We cloned most, if not all, of the aflatoxin pathway genes. A consensus gene cluster for aflatoxin biosynthesis was discovered in 2005. The factors that affect aflatoxin formation have been studied. In this report, the author summarize...
متن کاملInhibition of aflatoxin biosynthesis by phenolic compounds.
The phenolic compounds acetosyringone, syringaldehyde and sinapinic acid inhibited the biosynthesis of aflatoxin B1 (AFB1) by A. flavus. Acetosyringone was the most active among the three compounds, inhibiting aflatoxin level by 82% at 2 m moll-1. The synthesis and accumulation of norsolorinic acid, an aflatoxin biosynthetic intermediate, was also inhibited. These results suggest that at least ...
متن کاملIdentification of averantin as an aflatoxin B1 precursor: placement in the biosynthetic pathway.
A new blocked mutant of Aspergillus parasiticus produces no detectable aflatoxin B1, but accumulates several polyhydroxyanthraquinones. One of these pigments was identified as averantin. This is the first report of its formation by A. parasiticus. Radiotracer studies with [14C]averantin showed that 15.3% of label from averantin was incorporated into aflatoxin B1. This incorporation was blocked ...

Five strains ofAspergillusfJavus lacking the ability to produce aflatoxins were examined in greenhouse tests for the ability to prevent a toxigenic strain from contaminating developing cottonseed with aflatoxins. All atoxigenic strains reduced contamination when inoculated into developing bolls 24 h prior to the toxigenic strain. However, only one strain, AF36, was highly effective when inocula...
